摘    要:目的:建立HPLC测定加味龟鹿二仙膏中甜菜碱含量的方法。方法:采用薄层色谱法鉴别加味龟鹿二仙膏的龟甲、甘氨酸;采用HPLC对制剂中甜菜碱进行含量测定,色谱柱:GL Inertsil ODS-SP色谱柱(150mm×4.6mm,5μm);柱温:25℃;检测波长:196nm;流动相:乙腈-0.1%三乙胺溶液(磷酸调PH值至7)(80:20);流速:0.8 mL/min。结果:龟甲、甘氨酸的薄层鉴别斑点清晰、专属性强;甜菜碱在10.4606~104.6064μg之间呈良好的线性关系(r=0.9997,n=6),平均回收率为97.57%,RSD%为1.41%(n=9)。结论:所用方法操作简单、重现性好、灵敏度高、结果准确可靠,能有效控制加味龟鹿二仙膏的质量。

Abstract:Objective: To establish a methed for content determination of betaine HPLC modified Guilu Erxian Gao. Methods: TLC method of modified Guilu Erxian Gao shell, glycine; the HPLC content determination of betaine in preparation, chromatographic column: GL Inertsil ODS-SP column (150mm×4.6mm, 5μm); column temperature: 25℃; the detection wavelength: 196nm; mobile phase acetonitrile triethylamine solution: -0.1% three (phosphoric acid to adjust the pH to 7) (80:20); flow rate: 0.8 mL/min. Result: the TLC spots tortoise shell, glycine clear, strong specificity; Betaine in 10.4606-104.6064μ showed a good linear relationship between G (r = 0.9997, n=6), the average recovery was 97.57%, RSD% was 1.41% (n=9). Conclusion: the method has the advantages of simple operation, good reproducibihty, high sensitivity,accurate and relible, and can effectively control the quality of modified Guilu Erxian Gao.
