益母草水苏碱对大鼠心肌细胞肥大的肌浆网钙摄取能力的影响

摘    要:目的:探讨益母草水苏碱(stachydrine,Sta)对去甲肾上腺素(norepinephrine,NE)诱导的新生大鼠心肌细胞肥大肌浆网钙摄取能力的影响。方法:采用不同浓度的益母草水苏碱(10-4、10-5、10-6mol/L)对NE诱导的心肌肥大模型组进行干预,通过大鼠心肌肌浆网的蛋白水平和钙摄取能力来反映益母草水苏碱对NE诱导的新生大鼠心肌细胞肥大的影响。结果:益母草水苏碱能够明显减少心肌细胞蛋白含量,降低Protein/DNA比值;益母草水苏碱可剂量依赖地提高NE诱导的心肌肥大细胞的肌浆网钙摄取功能。结论:益母草水苏碱对NE诱导的心肌肥大细胞的肌浆网钙摄取功能有一定的程度的提高,与剂量有一定的依赖关系,随着益母草水苏碱的浓度的提高,肌浆网的钙摄取速率也明显提高。

Abstract:Objective:To investigate the Stachydrine (Sta) on norepinephrine (NE)-induced hypertrophy of neonatal rat cardiac myocyte sarcoplasmic reticulum calcium uptake capacity. Methods:different concentrations of Stachydrine (10-4,10-5,10-6mol/L) on the NE-induced cardiac hypertrophy model group intervention,by rat cardiac sarcoplasmic reticulum calcium uptake of protein levels and the ability to Stachydrine reflect on NE-induced hypertrophy of neonatal rat cardiac myocytes studied. Results:Stachydrine can significantly reduce the protein content of myocardial cells and reduce Protein / DNA ratio; Stachydrine in a dose-dependent increase NE-induced cardiac mast cells in sarcoplasmic reticulum calcium uptake. Conclusion:Stachydrine on NE-induced cardiac mast cells in sarcoplasmic reticulum calcium uptake have a certain degree of increased dose-dependent relationship between certain,with the concentration of Stachydrine improvement of sarcoplasmic reticulum calcium uptake rate has improved markedly.
