补肾通窍方对糖尿病大鼠下丘脑室旁核神经元Bax与Bcl-2表达的影响

摘    要:目的:探讨补肾通窍方对糖尿病大鼠下丘脑室旁核神经元Bax与Bcl-2表达的影响。方法:腹腔内注射链脲佐菌素(streptozotocin,STZ)55mg/kg制作糖尿病大鼠模型,采用免疫组化SABC法检测各组大鼠下丘脑室旁核神经元Bax与Bcl-2的表达。结果:①糖尿病模型组Bax阳性细胞数较正常对照组增多(P〈0.01),Bcl-2阳性细胞数减少(P〈0.01)。②补肾通窍方治疗组和胰岛素治疗组Bax阳性细胞数比糖尿病模型组减少(P〈0.01),而Bcl-2阳性细胞数则增多(P〈0.01)。③补肾通窍方治疗组和胰岛素治疗组的Bax和Bcl-2阳性细胞数无显著差异(P〉0.05)。结论:补肾通窍方能抑制糖尿病大鼠下丘脑室旁核神经元Bax的表达,促进Bcl-2的表达,与胰岛素相比哪者效果更好尚待研究。

Abstract:Objective:To observe the effects of Bushen Tongqiao decoction on expressions of Bax and Bcl-2 in paraventricular hypothalamic nucleus neurons of diabetic rats.Methods:The diabetic rat models were induced by intraperitioneal injection of streptozotocin(STZ).The expression of Bax and Bcl-2 was detected by immuno-histochemistry SABC.Results: ①Compared with normal control group,the positive cell's density of the expressions of Bax increased in paraventricular hypothalamic nucleus neurons of diabetic mellitus control group(P0.01),while the positive cell's density of the expressions of Bcl-2 decreased(P 0.01).②Compared with diabetic mellitus control group,the positive cell's density of the expressions of Bax decreased in paraventricular hypothalamic nucleus neurons of Bushen Tongqiao decoction prevention and cure group and insulin prevention and cure group(P0.01),while the positive cell's density of the expressions of Bcl-2 increased(P0.01).③Compared with insulin prevention and cure group,the positive cell's density of the expressions of Bax and Bcl-2 have no obvious difference in paraventricular hypothalamic nucleus neurons of western control group(P0.05).Conclution: Bushen Tongqiao decoction and insulin both can restrain the expressions of Bax which in paraventricular hypothalamic nucleus neurons of diabetic rats,and advance the expressions of Bcl-2.But which is more effective need further research.
