地骨皮调控H2O2所致内皮细胞凋亡的有效部位及相关信号通路研究

摘    要:目的:筛选地骨皮调控H2O2所致人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HU-VEC)凋亡的有效部位,并研究相关的信号转导通路,探讨其保护作用的分子机制。方法:用H2O2构建凋亡模型,运用MTS检测细胞存活率,流式细胞仪检测凋亡率筛选地骨皮有效部位,及Western观察相关信号通路蛋白P-AKT、P-NF-KB表达情况。结果:地骨皮30%,50%,70%醉提取部位均可以减少HUVEC的凋亡,可以明显增加P-AKT的表达,降低P-NF-KB活性,尤其55%醇提地骨皮用量最小效果最佳。结论:地骨皮抑制H2O2所致HUVEC凋亡的主要成分在55%醇提部位,它可以通过AKT、NF-κB信号通路起到抗凋亡的作用。

Abstract:Objective: This study was performed to select the most effective site of Cortex Lycii Radicis that protect human umbilical vein endothelial cells(HUVEC) from H2O2-induced apoptosis and study the Associated signaling pathway to reveale the possible molecular mechanisms involved.Methods: Apoptosis model of HUVEC was established by inducing of H2O2.The apoptosis cells were assessed by MTS and Annexin V-propidium iodide double staining flow cytometry to select the effective site of CLR against H2O2-induced apoptosis.The activities of AKT NF-KB were assayed by Western blot to investigate the underlying molecular mechanisms.Results:Stimulated with H2O2.a high fraction of HUVEC underwent apoptosis.While in the presence of CLR(30%,50% and 70% alcohol extracts).HUVEC apoptosis was blocked.The activation of AKT induced by H2O2 was significantly improved and the activation of NF-κB was significantly reduced,55% alcohol extract from CLR seemed to be more effective than 35% and 75% alcohol extract to protects HUVEC from H2O2-induced apoptosis.Conclusion: The 55% alcohol extract is the most effective site of CLR to protect HUVEC from H2O2-induced apoptosis through inhibition of the activation of AKT and NF-KB.
