蝉花总多糖对细胞免疫和体液免疫反应的促进作用

摘    要:目的:研究蝉花总多糖(CCP)体内外对细胞免疫和体液免疫的影响。方法:首先用MTT法和ELISA法测定小鼠脾细胞增殖和IgG抗体的生成水平,以研究CCP体外对细胞免疫和体液免疫的影响;然后通过DNFB诱导的小鼠迟发型超敏(DTH)反应和绵羊红细胞(SRBC)诱导的小鼠血清抗体生成实验进一步观察CCP体内对特异性细胞免疫和体液免疫反应的影响。结果:CCP 0.16~100μg/mL各浓度不仅能直接刺激小鼠脾细胞的增殖和IgG抗体生成,对Con A或LPS诱导的小鼠T、B淋巴细胞增殖和IgG抗体生成也具有显著的促进作用,并呈现良好的浓度依赖性。CPP 20 mg/kg可显著提高小鼠的DTH反应;10和20 mg/kg可剂量依赖性地显著提高抗SRBC抗体的生成。结论:CCP在体内外均能有效促进细胞免疫和体液免疫反应。

Abstract:Objective: To investigate the effects of polysaccharides from Cordyceps cicadae(CCP) on the cellular and humoral immune responses in vitro and in vivo.Methods: The effects of CCP on the cellular and humoral immune responses in vitro were studied by spleocyte proliferation(MTT assay) and IgG production(ELISA assay),respectively.The effects of CCP on the cellular and humoral immune responses in vivo were observed by DNFB-induced delayed type hypersensitivity(DTH) and sheep red blood cell(SRBC)-induced antibody production in mice.Results: CCP(0.16~100 μg/mL) not only directly stimulated the proliferation and IgG production of mice splenocyte,but also promoted the Con A or LPS-stimulated proliferation activity and IgG production of mice splenocytes in concentration-dependent manner.CCP remarkably promoted the DTH reaction at the dose of 20 mg/kg and enhance anti-SRBC antibody titers in naive mice at the dose of 10 and 20 mg/kg.Conclusion: CCP has marked promotive effect on the cellular and humoral immune responses in vitro and in vivo.
