电针干预高血压大鼠脑缺血Ang1 TSP-1的表达及细胞超微结构的研究

摘    要:目的:观察电针对易卒中型肾血管性高血压大鼠(RHRSP)脑缺血-再灌注后不同时间点促血管生成素-1(Ang 1)、凝血酶敏感蛋白-1(TSP-1)及超微结构表达影响。方法:用环形银夹使SD大鼠的双侧肾动脉狭窄,制成RHRSP,再用线栓法制成一侧大脑中动脉闭塞(MCAO)模型,大鼠随机分为空白组、假手术组、模型组、电针组。运用免疫组织化学法和光镜及电镜观察电针对脑缺血2h后再灌注1、7、14、28天大鼠脑内缺血区Ang 1、TSP-1表达的干预作用,并与模型组比较。结果:空白组、假手术组没有Ang 1、TSP-1阳性细胞表达,Ang 1在脑缺血2h后再灌注7天明显增多,14天达高峰,28天仍高于正常组水平;电针组在7、14、28天且较模型组升高更加明显,差异有统计学意义(P〈0.05)。模型组在1天后出现TSP-1阳性细胞,7天达到高峰并延续到14天,28天则明显减弱;电针治疗组在1、7、14天阳性细胞数较模型组明显减少(P〈0.05)。超微结构发现电针组微血管内皮细胞在各时间点损伤较模型组轻。结论:电针对RHRSP脑缺血-再灌注损伤的保护作用,可能与上调Ang 1及下调TSP-1的表达,促进脑缺血区的血管的重建有关。

Abstract:Objective:To observe the effects of the electric acupuncture on the expression of angiopoietin-1(Ang 1) thrombospodin-1(TSP-1) and ultramicrostructure during the different time of the cerebral ischemia and reperfusion in Stroke-prone-Renovascular-Hypertensive-Rats(RHRSP).Methods:Stroke-prone-Renovascular-Hypertensive-Rats(RHRSP)models were established by bilaterally narrowing the renal artery with sliver loop clips and the models of rat middle cerebral artery occlusion(MCAO) were created by stringing middle cerebral artery occlusion in RHRSP.2-hour ischemia later,the models were randomly divided into 4 groups : the normal,sham group,model group and electric acupuncture group.The effects of the electric acupuncture on the expression of Ang 1 TSP-1 and ultramicrostructure in the rat cerebral ischemic tissue were detected by immunohistochemistry,light microscope and electron microscope.After 1-day,7-day,14-day and 28-day reperfusion,which would be compared with the model g control group.Results:There were null-positive Ang 1 TSP-1 cells of expression in the normal and sham group,sham-operated group,The positive Ang 1 cells were found after 2-hour ischemic and 1-day reperfusion,which was increased after 14-day.and reduced after 14-day significantly after 28-day.Contrast to the model group,the expression of The positive Ang 1 cells in the electric acupuncture group were obviously increased at 7-day,14-day and 28-day ischemic and reperfusion(P0.05);The expression of positive TSP-1 cells in model group were found area after 2-hour ischemia and 1-day reperfusion.It was increased significantly after 7-day reperfusion and up to the maximum after14-day reperfusion;reduced after 28-day.Contrast to the model group,the numbers of positive TSP-1 cell in electric acupuncture group were decreased after 1-day 7-day and 14-day ischemic and reperfusion(P0.05).Conclusion:The electric acupuncture can promote the neurofunctional recovery in cerebral ischemic area.The mechanism may be related to up-regulating the expression of Ang 1 and down-regulating TSP-1 and rebuilding the micovascular net of cerebral ischemic tissue.
