中药怀牛膝的液相色谱指纹图谱研究

摘    要:目的:建立怀牛膝药材液相色谱指纹图谱检测方法,为其质量控制提供方法。方法:以蜕皮甾酮为参照物,采用RP-HPLC法,色谱柱为Kromasil-C18(250mm×4.6mm,5μm),流动相为乙腈-0.4%磷酸水溶液梯度洗脱,检测波长为254nm。结果:该方法可使怀牛膝药材中各成分较好分离,并根据检测结果确定了14个共有指纹峰,建立了怀牛膝HPLC指纹图谱共有模式。结论:建立的指纹图谱方法简便可靠,可以作为怀牛膝药材质控的重要方法。

Abstract:Objective: To establish detecting methods with fingerprint chromatogram for the Achyranthes bidentata. Methods: Kromasil- 5 C18 column ( 250 mm ×4.6 mm , 5μm ) was used with a mixture of acetonitrile- 0.4% phosphorus acid as mobile phase in a gradient mode, and with ecdysteron as the reference substance. The wavelength was 254nm. The data were analyzed with' Computer Aided Similarity Evaluation' software. Results: Established the typical fingerprint chromatogram. Conclusion: The method is simple and accurate with a good reproducibility and can be well used in quality control of Achyranthes bidentata.
