基于MARX发生器的电穿孔经皮给药仪的设计

目的:研制一种新型的药物促渗仪器,该仪器可产生瞬态高压脉冲,使皮肤发生电穿孔,从而促进药物尤其是大分子药物的经皮渗透量,对治疗癌症等疾病有着积极的意义。方法:运用MARX发生器原理,以IGBT为开关,设计三级MARX电路,同时配合微控制器AT89S52控制输出脉冲的大小、频率、脉冲数和参数显示。结果:该仪器产生出电压0～400 V、脉宽μs级至ms级可调的指数衰减波。使用不同参数的脉冲将穿心莲提取物作用于家兔和小白鼠的离体皮肤上,穿心莲提取物的累积经皮渗透量大于被动扩散条件。结论:基于MARX发生器的电穿孔仪能产生瞬态高压脉冲,这种脉冲可提高药物经皮渗透的渗透量,可为提高用于癌症治疗的大分子药物在人体内的利用率提供新途径。

Design of Electroporation Transdermal Drug Delivery Device Based on MARX Generator

Objective To develop a new drug penetration enhancers instrument to produce high voltage transient pulse that can bring the skin about electroporation and consequently promote drugs percutaneous permeation amounts especially for the macromolecules drug,which has a positive meaning to treatment of diseases such as cancer.Methods With the principle of MARX generator,three steps MARX circuit was designed using IGBT as switch,and microcontroller AT89S52 was also used in conjunction to control the size,frequency,pulse number of the output pulse and parameter display.Results The instrument produced a voltage of 0 to 400 V,pulse width adjustable from μs level to ms level index attenuation wave.Using pulses with different parameters,andrographis paniculata extract was acted on the isolated skin of the rabbit and the mouse,the cumulative percutaneous permeation amounts of which was higher than that of passive diffusion.Conclusion Electroporation transdermal drug delivery device based on MARX generator can produce high voltage transient pulse,which can improve the percutaneous penetration amounts of drug,and provide a new way for improving utilization of large molecules such as cancer drugs in the body.Chinese Medical Equipment Journal,2012,33（6）：18-20]
