气电纺纳米羟基磷灰石-聚羟基丁酸酯复合纤维支架与大鼠成骨细胞体外生物相容性研究

摘    要:目的研究气电纺纳米羟基磷灰石-聚羟基丁酸酯[nanosized hydroxyapatite—poly(3-hydroxybutyrate),nHAP-PHB]复合纤维支架对大鼠成骨细胞体外粘附、增殖及骨钙素分泌活性的影响。方法实验组将大鼠成骨细胞接种于气电纺nHAP—PHB复合纤维支架进行体外培养,对照组将大鼠成骨细胞接种于气电纺纯聚羟基丁酸酯(PHB)纤维支架行体外培养。2组进行扫描电镜观察、四甲基偶氮唑盐比色检测及骨钙素检测,评价nHAP—PHB复合纤维支架对大鼠成骨细胞体外粘附、增殖以及骨钙素合成分泌活性的影响。结果扫描电镜观察结果显示接种4h后,与纯PHB纤维支架相比,细胞在nHAP—PHB复合纤维支架上的铺展更完全。培养后第3、7、10、14天,实验组细胞增殖活性均强于对照组(P〈0.05),骨钙素分泌活性均高于对照组(P〈0.05)。结论nHAP—PHB复合纤维中nHAP增强了支架材料与大鼠成骨细胞的生物相容性。
