中国林蛙卵油气相色谱数字化指纹图谱和定量鉴别研究

摘    要:目的建立中国林蛙卵油气相色谱(GC)数字化指纹图谱。方法采用气相色谱法,FFAP(30m×0.25mm×0.33μm),氢焰离子化检测器(FID),进样量1.0μL。采用程序升温:120℃(1min)→20℃·min^-1→220℃(54min)。以“中药色谱指纹图谱超信息特征数字化评价系统”软件进行数字化和定量评价。结果以正二十四烷峰为参照物峰,确定20个共有峰,建立了中国林蛙卵油GC数字化指纹图谱。获得了以色谱指纹图谱指数F等42参数对中国林蛙卵油GC指纹图谱的超信息特征数字化评价结果和系统指纹定量法定量鉴别结果。结论该气相色谱数字化指纹图谱具有较好的精密度和重现性,适用于中国林蛙卵油质量控制。

Abstract:Objective To develop the GC digitized fingerprint of Forest Frogs Oviduct. Methods Chromatographic fingerprints were obtained by injecting 1.0 μL sample solution each time on an FFAP (30 m×0.25 mm×0. 33 μm) with column temperature from 120 ℃ (1 min) to 220℃ (54 rain) with 20 ℃ · min ^-1. FID was used as the detector and n-Tetracosane was used as referential standard. The chromatographic fingerprints were entirely evaluated by the soft- ware of the digitized evaluation system of Traditional Chinese Medicine fingerprint with the super information characteristics. Results Twenty co possessing peaks were selected as the fingerprint peaks of Forest Frogs Oviduct and the similarities of the chromatographic fingerprints were calculated by taking n-Tetracosane peak as the referential peak. The chromatographic fingerprints were charateristically digitized and evaluated by the software of the digitized evaluation system of Traditional Chinese Medicine fingerprint with the super information characteristics to provide 42 parameters such as chromatographic fingerprint index (F), chromatographic fingerprint resolution index (RF), which can thoroughly disclose the hidden information in the fingerprints. The quality grade resluts of Forest Frogs Oviduct were identified by the system fingerprint quantified method. Conclusion This digitized fingerprint method with good precision and reproducibility can be applied in the quality control of Forest Frogs Oviduct.
