摘    要:目的 探讨环状核糖核酸中hsa-circ-0015382通过调控miR-330-3p/SP4轴参与妊娠期孕妇子痫前期的发生。 方法 取子痫前期孕妇及正常孕妇顺产胎盘组织和产前静脉血提取总RNA。采用qRT-PCR检测hsa-circ-0015382和miR-330-3p的表达。采用Western Blotting检测特异性蛋白4(Specificity protein 4,SP4)的表达。原代培养子痫前期产妇胎盘滋养层细胞(PPC),采用双荧光素酶报告基因检测SP4是miR-330-3p的直接靶基因。采用MTT法检测hsa-circ-0015382和miR-330-3p对胎盘滋养层细胞增殖能力的影响,用Transwell法和细胞划痕实验检测hsa-circ-0015382和miR-330-3p对PPC细胞侵袭迁移能力的影响。 结果 hsa-circ-0015382在子痫前期孕妇胎盘组织和血浆中异常高表达,SP4是miR-330-3p在PPC细胞中的直接靶基因。在PPC细胞中抑制hsa-circ-0015382表达后,miR-330-3p的表达升高,hsa-circ-0015382和SP4的表达显著降低,同时细胞增殖能力和侵袭迁移能力增强,差异具有统计学意义(P<0.05)。在PPC细胞中转染miR-330-3p mimics后,细胞增殖能力和侵袭迁移能力增强,差异具有统计学意义(P<0.05)。 结论 hsa-circ-0015382通过调控miR-330-3p/SP4轴参与子痫前期的发生。

Abstract:Abstract: Objective To investigate the involvement of hsa - circ - 0015382 in the occurrence of preeclampsia in pregnant women by regulating the miR - 330 - 3p /SP4 axis. Methods Total RNA was extracted from the eutocia placental tissues and prenatal vein blood samples of pregnant women with preeclampsia and normal pregnant women. qRT - PCR was used to detect the expression of hsa - circ - 0015382 and miR - 330 - 3p. Western Blotting was used to exam the expression of Specificity Protein 4 ( SP4) . The placental trophoblast cells of preeclampsia ( PPC) were cultured primarily. The dual luciferase reporter gene assay was adopted to exam whether SP4 was the direct target gene of miR - 330 - 3p. MTT method was used for detecting the effects of hsa - circ - 0015382 and miR - 330 - 3p on the proliferation of PPC, and Transwell method and cell wound scratch assay were used to test the influences of hsa - circ - 0015382 and miR - 330 - 3p on the invasion and migration of PPC. Results hsa - circ - 0015382 was abnormally highly expressed in placental tissue and plasma of pregnant women with preeclampsia. SP4 was the direct target gene of miR - 330 - 3p in placental trophoblastic cells of preeclampsia ( PPC) . After inhibiting the expression of hsa - circ - 0015382 in PPC, the expression of miR - 330 - 3p was increased, while the expression of hsa - circ - 0015382 and SP4 was decreased. Meanwhile, proliferation and invasion and migration capabilities were enhanced, and the difference was statistically significant ( P < 0. 05) . After transfecting miR - 330 - 3p mimics into PPC, the proliferation and invasion and migration capabilities were enhanced, and the difference was statistically significant ( P < 0. 05). Conclusion hsa - circ - 0015382 participates in the occurrence of preeclampsia by regulating the miR - 330 - 3p /SP4axis.
