局灶性脑缺血大鼠海马CA_3区突触体素动态表达

摘    要:目的 研究局灶性脑缺血后海马CA3 区突触体素的动态表达以及神经修复的可塑性。方法 选取健康成年SD大鼠 4 0只 ,随机分为脑缺血组和对照组 ,每组又分为术后 7、14、2 1、30天 4个时间点 (n =5 )。采用线栓法建立大脑中动脉脑缺血大鼠模型 ,应用免疫组化技术观察海马CA3 区突触体素的表达。结果 脑缺血后各时间段突触体素阳性反应物表达的光密度较对照组明显降低 (P <0 .0 1) ;缺血后 7天至 2 1天突触体素表达逐渐回升 (P <0 .0 1) ,缺血后 30天又下降 ,但仍低于对照组。结论 突触体素可作为海马CA3 区神经元功能和损伤修复的标示物。

Abstract:Objective To study the expression of synaptophysin in hippocampus at different times after a focal cerebral ischemia,and the plasticity of neural repair in hippocampus after injury. Methods 40 healthy adult SD rats were divided randomly into experimental and control group at 7, 14, 21 and 30 days(n=5 for time point) after surgery. Unilateral cerebral ischemia was induced in rats by permanent occlusion of the distal middle cerebral artery. The expression of synaptophysin was examined by immunohistochemical technique. Results The optical density of synaptophysin immunoreactivity in CA 3 area of hippocampus of ischemic animals decreased significantly compared with sham controls. The optical density of positive products of synaptophysin increased in experimental group gradually from days 7 to 21 postoperatively, but still much lower than those in sham controls(P<0.01). Conclusion Synaptophisin might be the marker for the function of neurons and repairment of the damaged neurons in CA 3 area of hippocampus.
