野生型p53基因转染对宫颈癌HeLa细胞生长及药物敏感性的影响

摘    要:目的 探讨转染外源野生型 p5 3基因对宫颈癌 He L a细胞生长的抑制作用 ,并了解 p5 3基因与He L a细胞对顺铂敏感性之间的关系。方法 利用脂质体介导 ,将含人野生型 p5 3c DNA的 p CB6 .p5 3质粒导入人宫颈癌 He L a细胞株中 ,筛选阳性克隆 ,用免疫组化 ABC法检测 p5 3基因的表达情况。加入顺铂 72小时后 ,用四唑盐比色试验检测存活的 He L a细胞数。结果 在 G418选择培养基中培养两周后 ,成功地生长出阳性细胞克隆。免疫组化法证实外源性 p5 3基因在瘤细胞中稳定存在并传给子代 ,p5 3基因转染对 He L a细胞的生长有明显的抑制作用。p5 3表达阳性的 He L a细胞对顺铂的敏感性明显增强。结论 野生型 p5 3基因转染能使 He L a细胞出现生长抑制和化疗敏感性增强

Abstract:Objective To investigate the effects of human wild type p53 gene transfection on the growth and chemosensitivity of human cervical carcinoma. Methods Recombinant eukaryotic expression vector pCB6.p53 containing human wild type p53 cDNA was introduced by lipofectamine transfection reagent into HeLa cell line. The expression of p53 was detected by immunohistochemistry. Cell proliferation was measured by methyl thiazolyl tetrazolium after the treatment of cisplatin. Results Expression of p53 was detected by immunohistochemistry. The growth rate of p53 transfected HeLa decreased. Wild type p53 positive HeLa was more sensitive to cisplatin, compared with the control cell lines. Conclusion The exogenous wild type p53 expression not only induced great suppression of cell growth but also increased chemosensitivity of human cervical carcinoma.
