VitE抗大鼠肝纤维化的实验研究

摘    要:目的研究VitE预防和治疗肝纤维化的作用。方法采用40%CCl4制备大鼠肝纤维化模型,观察VitE对大鼠氧化及肝纤维化各项指标的影响。结果VitE能显著降低大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、Ⅲ型前胶原(PCⅢ)及肝组织中脂质过氧化物丙二醛(MDA);提高肝组织中超氧化物歧化酶(SOD)活性;并能显著减轻肝脏胶原纤维增生程度。结论VitE具有抗肝纤维化作用。

Abstract:Obiective The aim of this study was to inquire into the effects of vitamin Eon antihepatic fibrosis. ANthods Thirty - six adult Wistar rats were randomly divided intothree groups (12rats/group): heathly control group, CCl4 injured rats left untreated (CCl4group), and CCl4 injured rats treated with vitamin E(Vitamin E group). Liver fibrosis wasinduced in rats by subcutaneous injection of 40% CCl4 oily solution. Vitamin E group wastreated with dietary supplementation of vitamin E(25Omg/kg). After 8 - week treatment, ratswere killed and the following parameters of hepatic fibrosis and antioxidation weredetermined: hepatic maiondialdehyde (MDA) and superoxidedismutase(SOD) levels, serumalanine minotransferase (ALT) aspartate aminotransferase (AST), hyaluronic acid(HA)precollagen type III(PCIII) and vit E concentrations, and histological staining of hepatictissues. ResuIts The results indicated the levels of serum ALT. AST. HA and PCIII in rats ofvitamin E group was significantly lower than that of the CCl4 group (P<0.05); in hepatictissue, the amount of MDA was decreased, the activities of SOD were significantly elevated,the proliferation degree of collagenous fibers was lower in vitamin E group than that of CCl4group. ConcIuhon The parameters indicate that vitamin E significantly protects againsthepatic fibrogenesis in the rats.
