细胞外基质蛋白介导晶体上皮细胞粘附的作用

摘    要:目的探讨纤粘连蛋白、层粘连蛋白、I型胶原3种细胞外基质蛋白促牛晶体上皮细胞(LEC)的粘附作用,以阐明后囊膜混浊(PCO)发生的组织病理学机制。方法应用3H-TDR掺入法研究纤粘连蛋白、层粘连蛋白、I型胶原和鸡卵白蛋白对体外培养的牛LEC的粘附作用。结果牛LEC能够粘附到包被有不同浓度的料粘连蛋白、层粘连蛋白、I型胶原培养板表面上,呈现出浓度和时间效应特点肥牛LEC不能够粘附到包被有不同浓度的鸡卵白蛋白培养板表面上。给哈构成晶体囊膜的层粘连蛋白和I型胶原,以及白内障摘除术后进入眼房水的纤粘连蛋白能够介导LEC粘附到后囊膜上,在PCO发生过程中起到重要的促进作用。

Abstract:Objective To inquire into the role of fibronectin, laminin and type I collagen in promotion of bovine lens epithelial cells adhesion and to explain the pathogenesis of posterior capsular opacification- Methods The 3H - TDR incorporating assays were applied to study adhesion of bovine lens epithelial cells cultured in response of fibronectin, laminin, type I collagen and ovalbumin in vitro. Results Bovine lens epithelial cells adhered to surfaces coated with varying concentrations of fibronectin, laminin and type I collagen, and it was shown to be dependent on concentration and time. Bovine lens epithelial cells did not adhere to ovalbumin- coated surfaces at any concentration tested. Conclusion The results of these studies indicate that adhesion of lens epithelial cells occurs in response to lens capsular protein type I collagen, laminin and fibronectin that is introduced into the aqueous humor after extracapsular cataract extraction, they may play a critical promotive role in the occurrence of posterior capsular opacification.
