GC-MSn同时测定小鼠血浆中冰片和尼莫地平质量浓度

 目的建立测定小鼠血浆中冰片和尼莫地平的气相色谱-质谱(GC-MSn)定量分析方法。分析两药合用后尼莫地平在小鼠体内的药动学变化。方法血浆样品经乙酸乙酯提取,采用选择性离子抽提和二级MS/MS质谱分别监测冰片和尼莫地平的血药浓度。结果本法可同时测定血浆中2种相对分子质量差异较大的药物质量浓度,冰片在0.01～10 mg·L^-1,尼莫地平在0.01～0.5 mg·L^-1内,峰面积与其质量浓度呈良好的线性关系。最低检测限分别为5.0,2.5 mg·L^-1,日内RSD分别为4.6%～6.7%,2.5%～5.2%;日间RSD分别为2.3%～9.9%,5.1%～6.6%(n=5)。结论该方法灵敏,简便,准确,适用于冰片和尼莫地平配伍应用的药动学研究。尼莫地平加冰片合用的动力学研究表明,冰片可以改变尼莫地平在体内的分布过程。

Simultaneous Determination of Borneol and Nimodipine in Mice Plasma by GC-MSn

OBJECTIVE To establish a method for simultaneous determination of borneol and nimodipine in mice plasma by GC-MSⁿ.METHODS The plasma sample was extracted with ethyl acetate,borneol and nimodipine were determined by means of selected ion extraction and MS₂,respectively.RESULTS The concentrations of the different compounds were simultaneously determined.The calibration curves were linear in the range of 0.01～10 mg·L^-1 for borneol,0.01～0.5 mg·L^-1 for nimodipine,respectively.The limits of detection were 5.0 and 2.5 mg·L^-1,respectively.The RSDs of Intra-day validation were 4.6 %～6.7%,2.5%～5.2%,and the RSDs of inter-day validation were 2.3%～9.9%,5.1%～6.6%(n=5) for borned and nimodipine,respectively.CONCLUSION This methods is sensitive,simple,accurate,which could be used to determine simultaneously the concentrations of borneol and nimodipine in mice plasma and study their pharmacokinetics.
