肝星状细胞中Skp2表达及其对纤维化、增值及机制探讨

摘    要:目的 观察肝星状细胞中Skp2表达及其对纤维化的影响。方法 利用 siRNA 沉默HSC-T6细胞中Skp2基因的表达,CCK - 8 法检测细胞增殖,流式细胞术检测细胞凋亡, qRT - PCR检测Skp2、P27、Col I、Col III mRNA的表达,Western blot方法检测Skp2、P27、Caspase - 3、Survivn的蛋白表达水平。结果 与NC - Skp2组比较,siRNA - Skp2组Skp2 mRNA的相对表达水平显著降低,细胞的增殖能力受抑制,凋亡能力增强(P<0.05)。此外,与NC - Skp2组比较,siRNA - Skp2组P27 mRNA相对表达水平明显增加,Col I、Col III mRNA相对表达水平明显降低,差异有统计学差异(P<0.05);与NC - Skp2组比较,siRNA - Skp2组Skp2、Survivn蛋白水平明显降低,Caspase - 3、P27蛋白水平明显增加,均有统计学差异(P<0.05)结论 降低Skp2基因在HSC - T6细胞中的表达水平,可抑制细胞的增殖和纤维化。

Abstract:Objective To observe the expression of Skp2 in hepatic stellate cells and its effect on fibrosis. Methods siRNA was used to silence the expression of Skp2 in HSC-T6 cells. CCK-8 assay was used to detect cell proliferation. Flow cytometry was used to detect cell apoptosis. qRT-PCR was used to detect the expression of Skp2, P27, Col I and Col III mRNA. Protein expression of Skp2, P27, Caspase-3 and Survivn was detected by Western blot. Results Compared with NC-Skp2 group, the relative expression level of Skp2 mRNA in siRNA-Skp2 group was significantly reduced, cell proliferation was inhibited and apoptosis was enhanced(P<0.05). In addition, compared with NC-Skp2 group, the relative expression level of P27 mRNA in siRNA-Skp2 group was significantly increased, and the relative expression levels of Col I, Col III mRNA were significantly lower(P<0.05); compared with NC- Skp2 group, Skp2, Survivn protein levels in siRNA-Skp2 group decreased significantly, caspase-3, P27 protein levels were significantly increased, there were statistically significant differences(P<0.05). Conclusion Reduce the expression of Skp2 in HSC-T6 cells can inhibit the cells Proliferation and fibrosis.
