外源性神经生长因子对三叉神经感觉神经细胞凋亡的影响

摘    要:目的:探讨三叉神经下颌支损伤后外源性神经生长因子(NGF)对感觉神经元细胞凋亡及神经电生理的影响。方法:制作大鼠三叉神经下颌支离断硅胶管桥接动物模型,采用TUNEL法观察NGF局部应用三叉神经感觉核区神经元细胞凋亡的变化;采用肌电位仪观察三叉神经感觉核区感觉神经元电生理(动作电位潜伏期Lat,波幅Amp,传导速度CV)的变化。结果:外源性NGF治疗组三叉神经感觉核神经元细胞的凋亡数量明显少于单纯桥接吻合对照组。术后30d,NGF治疗组与单纯桥接吻合对照组相比,感觉神经元动作电位潜伏期短、波幅度、传导速度快。结论:外源性NGF在损伤局部应用,可在一定程度上抑制神经元细胞凋亡,促进神经电生理的恢复,从而对三叉神经损伤后神经的修复具有一定的促进作用。

Abstract:Objective To observe the effects of exogenous NGF on apoptosis of neurons and electro physiology of nerve after mandibular branch injury.Methods Established mandibular branch injuried model which anastomosed with silicone tube.Same dose of NGF or NS were filled into silicone tubes.To observe the change of apoptosis of sensory neurons and the change of action potential of neurons, TUNEL and electro physiological methods were applied respectively.Results The numbers of apopotosis sensory neurons of NGF group were significantly less than those of simply anastomosed group.The neurons action potential were significantly enhanced by exogenous NGF administrating in above mentioned models.Conclusion Exogenous NGF play protecting role in injured trigeminal nerve by inhibiting apoptosis of sensory neurons in injured area.
