肾细胞癌MMP-9、VEGF表达与MVD的相关性

摘    要:目的探讨基质金属蛋白酶-9(MMP-9)与血管内皮生长因子(VEGF)在肾癌组织中的表达及其与血管生成、肿瘤侵袭和转移的关系。方法采用免疫组织化学SP法,检测75例肾细胞癌、12例正常肾组织中MMP-9与VEGF的表达情况;CD34免疫组化染色方法标记微血管内皮计数微血管密度(MVD)。结果肾癌组织MMP-9与VEGF表达均明显高于正常肾组织(F=13.12、9.76;q=4.568~9.333;P〈0.05),不同病理类型的肾癌组织中MMP-9与VEGF表达差异均无显著性(q=0.035~2.209,P〉0.05),不同临床分期肾癌组织MMP-9与VEGF表达差异均有显著性(F=24.95,3.54;q=3.086-11.846;P〈0.05)。MVD与VEGF、MMP-9蛋白表达之间均呈显著正相关(r=0.381、0.375,P〈0.05),VEGF与MMP-9蛋白表达之间也有显著相关性(r=0.507,P〈0.05)。结论MMP-9、VEGF与肾细胞癌的血管生成有关,MMP-9与VEGF表达检测可作为反映肾细胞癌侵袭和转移潜能的生物学指标。

Abstract:Objective To study the expressions of MMP-9 and VEGF in renal carcinoma, and their correlation with angiogenesis, tumor invasion and metastasis. Methods Expressions of MMP-9 and VEGF were detected immunohistochemically in tissue from the carcinoma (RCC, n=75) and normal renal tissue (n=12). The MVD was estimated via counting the number of endothelium displayed by CD34 expression. Results Expressions of MMP-9 and VEGF in RCC were significantly higher than that of normal renal tissue (F=13.12;9.76;q=4.568-9.333;P<0.05), which were not related to pathological type (P>0.05), but related to clinical staging (F=24.95,3.54;q=3.086-11.846,P<0.05). A positive correlation between MVD, MMP-9 and VEGF was observed (r=0.381,0.375;P<0.05). Conclusion MMP-9 and VEGF correlate significantly with angiogenesis in RCC, suggesting of expressions of MMP-9 and VEGF may be biological indexes in detecting invasion and metastasis of RCC.
