摘 要: | 目的 观察正常人CIK(cytokine inducedkiller)细胞对人卵巢癌细胞株SKOV3ip的体外细胞毒活性以及对卵巢癌腹水瘤模型的体内抗肿瘤作用。方法 取正常人的外周血单个核细胞 (PBMC) ,加入细胞因子γ IFN、IL 2、抗 -CD3单抗和IL - 1,体外诱导成CIK细胞 ,用MTT法测定CIK细胞体外对人卵巢癌细胞株SKOV3ip的细胞毒活性 ,并与CD3AK作对比。在Babl/c裸小鼠腹腔内接种卵巢癌SKOV3ip细胞 ,观察CIK细胞对荷瘤鼠的抑瘤作用。结果 体外实验证明 ,CIK细胞对SKOV3ip有明显的细胞毒活性 ,对比CD3AK其抑瘤率为 89.7%与 6 7.1% (P <0 .0 5 )。裸鼠卵巢癌腹水瘤模型体内实验表明 ,CIK细胞能够显著抑制癌性腹水生长 ,其抑制率可达 10 0 %。结论 CIK细胞是一种新型、高效的免疫活性细胞 ,具有较强的体内外抗卵巢癌细胞活性 ,具有明显的抑制癌性腹水生长的作用 ,有可能用于临床上卵巢癌腹水的过继性免疫治疗

Anti-Tumor Effects of CIK Cells Against Ovarian Carcinoma Cell Lines SKOV3ip In Vitro and In Vivo |

Abstract: | Purpose To investigate the proliferation profile and the cytotoxicity of cytokine induced killer (CIK) cells from healthy donors in vitro and their anti tumor Affects in Balb/c nude mice bearing SKOV3ip cells strain of ascites tumor from ovarian. Methods Peripheral blood monouclear cells (PBMC) from healthy donors were induced to become CIK cells by cytokine in vitro incubation.The CIK cells were analyzed on different time points by FACS;Compared with CD3AK cells,the cytotoxicity in vitro by MTT assays and the antitumor activity of the CIK were evaluated in Balb/c nude mice bearing SKOV3ip cells from ovarian carcinoma. Results CIK cells possessed higher antitumor cytotoxic activity on SKOV3ip cells in vitro than CD3AK cells; In addition,CIK cells had a stronger suppressive effect on the tumor growth in Balb/c nude mice bearing SKOV3ip tumor compared with CD3AK cells in vivo median inhibitory rates 89.7 % vs 67.1%,( P <0.05). Conclusions CIK cells were highly efficient cytolytic effector cells which have a stronger significant suppression against growth of Ovarian carcinoma in vitro and in vivo .The result provides an experimental basis for CIK to clinical application as a adoptive immunotherapy. |
