缬沙坦对兔动脉粥样硬化血管VCAM-1表达的影响

摘    要:目的研究缬沙坦对动脉粥样硬化血管VCAM-1表达的影响。方法采用高胆固醇饲料喂饲另加牛血清白蛋白一次性注射方法,建立家兔动脉粥样硬化模型。30只纯种大白兔随机分为3组,A组为正常饮食组,B组为高脂饮食组,C组为高脂饮食加缬沙坦组。喂养8周后测定各组血清中总胆固醇(TC)、甘油三酯(TG)及低密度脂蛋白胆固醇(LDL-C),免疫组化观察内膜粥样硬化斑块中VCAM-1的表达情况。结果高脂饮食组、高脂饮食加缬沙坦组血清TC、TG、LDL-C水平较对照组显著升高(P<0.01),高脂饮食组、高脂饮食加缬沙坦组血清TC、TG、LDL-C水平差异无显著性。缬沙坦药物干预组内膜增生较高脂组明显减轻。VCAM-1在缬沙坦药物干预组中表达较高脂组明显减少。结论缬沙坦可明显抑制动脉粥样硬化的发生。其机制与减轻兔动脉粥样硬化血管VCAM-1表达,抑制动脉粥样硬化过程中的炎性反应密切相关。

Abstract:Objective To investigate the effects of losartan on expression of VCAM-1 in a rabbit model of atherosclerosis.Methods An atherosclerotic rabbit model was established by feeding high cholesterol diet supplemented by bovine serum albumin injection.The rabbits were randomly divided into three groups:normal food group,high cholesterol group,high cholesterol valsartan group,ten rabbits in each group.Blood samples were collected at the end of 8 weeks for examination of serum lipid levels and the expression of VCAM-1 were detected by immunohistochemistry.Results Rabbits fed with cholesterol-rich diet showed higher serum lipid levels(P<0.01) than those fed with normal diet.Serum lipid level of Rabbits fed with cholesterol-rich diet showed no difference.The ratio of lesion of intimal area of valsartan treated group reduced significantly than those of high cholesterol group(P<0.05).The expression of VCAM-1 of valsartan treated group decreased significantly than those of high cholesterol group.Conclutions Valsartan could inhibit atherosclerotic progression.The mechanism may be related to inhibition of inflammation and reduction in the expression of VCAM-1.
