摘    要: 目的:探讨β-羧乙基锗倍半氧化物(Ge-132)在体内清除自由基、抗氧化作用。方法:下列药物作用于烈酒造成氧应激态的大鼠,观察其肝线粒体超氧化物歧化酶活性和肝脏的病理变化,并与维生素C进行了比较。Wistar大鼠随机分为4组。正常对照(A),烈酒(B),烈酒+维生素C(C),烈酒+Ge-132(D)。各组大鼠常规饲喂。每种药的给药方法如下:烈酒(含体积分数为55%乙醇),5 g/(kg·d),从第8天开始ig共2周;维生素C 200 mg/(kg·d),从第1天开始sc共3周;Ge-132 200 mg/(kg·d),从第1天开始ig共3周。结果:肝线粒体铜锌超氧化物歧化酶活性,烈酒组与对照组比较,其活性下降(P<0.05),烈酒+Ge-132和烈酒+维生素C组均与烈酒组比较,其活性有上升趋势。肝脂肪变性在烈酒+Ge-132组较烈酒组为轻,烈酒+维生素C组病变不明显。结论:Ge-132在一定程度上保护肝线粒体铜锌超氧化物歧化酶活性,改善肝脂肪变性,但对肝脏病变的保护作用不如高浓度维生素C。

Abstract:OBJECTIVE:To examine the pharmacological effect of β-carboxyethyl germanium sesquioxide(Ge-132) on scavening free radical and the antioxidation in vivo.METHOD:The effects of the drugs on the rats under oxidative stress induced by strong wine were investigated by observing the activities of superoxide dismutase(SOD) in liver mitochondria and pathology of liver in the rats.Wistar rats were randomly divided into four groups(n=10):A:control;B:wine;C:wine+vitamin C;D:wine+Ge-132.The rats in all groups were fed with regular food.The dose and route of administration of drugs are as follows:Strong wine(55% ethanol),5 g/(kg·d),ig from 8 th day for 2 weeks.Vitamin C 200 mg/(kg·d) sc from 1 st day for 3 weeks.Ge-132 200 mg/(kg·d) ig from 1 st day for 3 weeks.RESULTS:Activity of Cu,Zn-SOD in liver mitochondria decreased in the strong wine group compared with the control(P<0.05),and tended to increase in the strong wine +Ge-132 group and wine+vitamin C group compared with the strong wine group.Liver steatosis was less in the strong wine+Ge-132 group than that in the strong wine group,but there was no significant change in the strong wine+vitamin C group.CONCLUSION:Ge-132 could protect from decreasing the activity of Cu,Zn-SOD in liver mitochondria to a certain extent and improve liver steatosis in strong wine rat.The effects of Ge-132 in improving fatty liver were not better than those of high concentration vitamin C.
