上皮性卵巢癌组织中Beclin1、PTEN的表达及其临床相关性研究

摘    要:目的探讨上皮性卵巢癌自噬基因Beclin1和PTEN蛋白的表达及其临床相关性.方法采用免疫组化方法检测25例正常卵巢组织、25例良性卵巢上皮性肿瘤、19例卵巢交界性肿瘤及69例卵巢上皮性癌组织中Beclin1及PTEN蛋白表达,比较四组组织中Beclin1及PTEN表达水平,以及与卵巢癌临床病理特征之间的关系.结果(1)Beclin1、PTEN蛋白在正常卵巢组织与良性卵巢肿瘤组织中表达较高,在交界性卵巢肿瘤组织中开始下降,在上皮性卵巢癌中表达最低(P〈0.05);(2)Ⅰ~Ⅱ期、高中分化、淋巴结无转移的卵巢癌组织中Beclin1、PTEN的表达高于Ⅲ~Ⅳ期、低分化、淋巴结有转移者,差异有统计学意义(P〈0.05).结论Beclin1、PTEN在上皮性卵巢癌中表达下调,可能与卵巢上皮性癌的发生、发展及临床预后有关.

Experssion of Beclin1 and PTEN Protein and Their Relationship with Clinic Characteristic of Epithelial Ovarian Cancer

Abstract:Objective To investigate the expression of Beclinl and PTEN protein in the epithelial ovarian carcinoma and their relationship with clinic characteristic. Methods The expression of Beclinl and PTEN protein in 25 cases of normal ovary, 25 cases of benign ovarian tumor, 19 eases of borderline ovarian tumor and 69 eases of epithelial ovarian cancer tissue were detected by immuohistochemical method. Results (1) The high expression of Beclinl and PTEN was found in normal and benign ovarian tumor tissues. The expression decreased in the borderline ovarian tissues, and was the lowest in the ovarian cancer (P 〈 0.05 ). (2) The expression of Beclinl and PTEN in stage Ⅰ-Ⅱ , high and middle grade and negative lymph node metastasis epithelial ovarian cancer were higher than that in stage Ⅲ-Ⅳ, low grade, positive lymph node metastasis ovarian cancer (P 〈0.05). Conclusion The expression of Beclinl and PTEN is down-regulated in the epithelial ovarian cancer. This may play an important role in the initiation and development of ovarian cancer.
