摘    要:目的 研究谷胱甘肽S转移酶系 (GSTs)基因多态性对非小细胞肺癌生存率的影响。方法 检测 5 70名非小细胞肺癌患者外周血淋巴细胞DNA上GSTs基因多态性。以回顾性队列研究的方法获得不同GSTs基因多态性对象肺癌治疗的生存结果 ,并用Kaplan Meier法和Cox回归分析探讨GSTs基因多态性对肺癌患者预后的影响。结果 接受化疗的肺癌患者GSTT1空白型基因与死亡的RR为 1.73,GSTT1和GSTM1均呈空白型基因的肺癌患者死亡的RR为 1.77,其中接受化疗的肺癌患者的RR为 2 .4 7。结论 GSTT1、GSTT1合并GSTM 1空白型基因与肺癌化疗后较低的生存率有关

Abstract:Purpose To study the association of glutathione S transferases (GSTs) GSTM1, GSTT1 and GSTP1 Polymorphisms with Survival of different therapies in non small cell lung cancers (NSCLCs). Methods A retrospective cohort study was used among 570 patients with NSCLCs. PCR was performed in detecting the polymorphisms of GSTT1, GSTM1, GSTP1 exon5 and GSTP1 exon6 from the genomic DNA of peripheral blood lymphocytes of the patients. Kaplan Meier method and Cox Regression was utilized for analyzing the relationship between GSTs polymorphisms and the death risks among NSCLCs patients with and without chemotherapies. Results GSTT1 null genotype had an increased RR (RR= 1.73 ) of death among patients with chemotherapies. GSTT1 null genotype combined GSTM1 null genotype had the significant RRs of death among all the patients and patients with chemotherapies. The RRs of death were 1.77 and 2.47 respectively. No significant survival influences on NSCLSc patients with different therapies were found from GSTP1 polymorphisms. Conclusions The polymorphisms of GSTT1?GSTT1 combined GSTM1 null genotype were associated with the higher risk of death among NSCLCs patients with chemotherapies.
