人血管内皮细胞的氧化损伤及药物保护作用的研究

摘    要:目的:研究盐酸曲美他嗪(万爽力)对过氧化氢(H2O2)体外诱导的人脐静脉内皮细胞(HUVEC)损伤的保护作用,并探讨其可能的作用机制。方法:在体外培养的人脐静脉内皮细胞上建立H2O2损伤模型,观察不同浓度的万爽力(1、10、100μmol/L)对细胞活力噻唑蓝、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性以及一氧化氮(NO)表达的影响。结果:万爽力可以提高H2O2诱导损伤的人脐静脉内皮细胞活性,降低MDA含量、增加SOD活性,促进NO的分泌,且呈一定的剂量相关性,差异具有统计学意义(P<0.05)。结论:万爽力对氧化损伤的内皮细胞具有保护作用,其作用机制可能与其抗脂质过氧化及清除超氧阴离子自由基有关。

Abstract:Objective To explore the protective effects of trimetazidine on the human umbilical vein endothelial cells (HUVECs) which were injured by hydrogen peroxide. Methods By using their primary culture in vitro, the HUVECs were injured by H2O2 and the cells were treated with three concetrations of trimetazidine(1,10 and 100 μmol/L).
