摘    要:目的研究人表皮移植至裸鼠皮下后表皮细胞的表型变化。方法将制备的人包皮表皮皮片移植到裸鼠背部皮下,7d取材。移植前后人表皮皮片标本行HE染色及细胞角蛋白10(CK10)、细胞角蛋白14(CK14)、p63免疫组织化学染色镜下观察。结果人表皮皮片移植到裸鼠皮下成活率达70%,移植成活的表皮基底层细胞增殖,细胞数增多,新生表皮增厚。免疫组化结果显示,与移植前相比,CK10表达减弱,CK14和p63表达增强,这与人皮肤创伤愈合过程中表皮细胞表型变化一致。结论移植人表皮到裸鼠皮下的方法可以模拟人皮肤创伤愈合过程,是一种可行的在体研究人表皮细胞的实验方法。

Abstract:Objective To explore the phenotype changing of human epidermis transplanted to subcutaneous area of athymic mouse. Methods The foreskin epidermis of human was transplanted to the subcutaneous area of athymic mice and taken out 7 days later. The sections of the epidermis were cut and studied with HE and immunohistochemical staining. Results The survival rate of the epidermic graft was about 70% ,the basal layer cells of survived epidermis grew and proliferated well and the number of cells increased. The results of immunohistochemical staining showed that the expression of cytokeratin 10 (CK10) decreased and expressions of cytokeratin 14 (CK14) and p63 increased after transplantation compared with pretransplantation, which was similar to the phenotype changes of the injured skin of human . Conclusion Transplantation of human epidermis to the subcutaneous area of athymic mouse simulates the healing process of human skin wound.
