HPLC测定8位肝癌病人5-FU血药浓度及其药物动力学研究

目的用改进的HPLC法测定8位肝癌病人静滴5-FU后7个时间点的血药浓度,对其进行药物动力学研究。方法采用HPLC-紫外检测5-FU血药浓度;并用3P97药动学程序计算出一系列药物动力学参数。结果血药浓度个体差异极大(2.5倍以上);5-FU药时曲线符合开放式二房室模型,AUC与年龄成正比。结论5-FU静脉滴注给药,血药浓度个体差异较大,临床效果(疗效、毒性)的差异也很大,故必须实施TDM(治疗药物监测),制订个体化给药方案,使用药更安全、有效、合理和经济,副作用更小。

Determination of 8 Hepatic Cancer's 5-FU Concentration in Serum by HPLC, and its Study Pharmakinetics

Objective To determine the concentration,and study the Phamacokinetics of 5-FU in serum at seven time points from 8 Hepatic Cancer`s by HPLC.Methods HPLC-UV was applied to determine the concentration of 5-FU in serum.A series of parameters were analysed by 3P97 Pharmacokinetics procedure.Result The concentration-time curve of 5-FU was fit to a TWO-compartment model.AUC was directly proportional to age.Individual difference was significant(above 2.5 multiple).Conclusions The individual difference of serum concentration was rather significant as well as clinical effect after 5-FU in administration.The TDM should be used for the individual administration,which will make the drug administration.To be more safe,effective,reasonable,economical and side effect less.
