摘    要:目的对本实验室保存的L1菌株进行He-Ne激光诱变,以获得一株耐高温且产酸能力强的菌株。方法运用He-Ne激光辐照L1菌株,对激光诱变参数进行优化,考察了突变株的耐温性、耐酸性和遗传稳定性。结果在照射剂量15min,照射功率10mW条件下,选育出一株耐高温且产酸能力强的菌株TL1,70℃培养下突变株菌数量是原菌株的12倍,菌数量最多达1.275×109个/mL,且其具有良好的遗传稳定性。结论He-Ne激光育种技术可以在高温乳酸菌中应用。

Abstract:Objective To screen the strain L1 with high thermortolerance and steady hydrogen--producing capability Through repeating He--Ne laser--induced breeding. Methods the strain Llwas irradiated by He-Ne laser to optimize the laser mutagenesis condition, and to test the thermortolerance,aciduric and the heredity.Results Using He--Ne lase (= 63218nm W= 10mW,T=15min) to irradiate the strain Ll,an mutant with thermortolerance and steady hydrogen--producing capability was obtained by the first, the second screened.The number of the strain TL1 improved 12 times than its former strain in 70℃ .The number of bacteria increased to 1.275 × 109U/mL.It has good heredity. Conclusion He-Ne laser mutagenesis could be used in the field of thermortolerant LAB.
