摘    要:目的:通过对登革病毒经垂直传播后亲代和子一代蚊体内病毒基因序列的分析,了解登革病毒在流行过程中的变异性。方法:用登革2型病毒(DEN-2)NGC株感染白纹伊蚊贵州兴义株,提取亲代及子一代蚊体内病毒总RNA,通过逆转录聚合酶链反应(RT-PCR)扩增蚊体内DEN-2病毒核酸,对阳性聚合酶链反应(PCR)产物进行序列测定和比较。结果:用DEN-2 NGC株感染白纹伊蚊贵州兴义株后,子一代蚊体内扩增的DEN-2 E基因区序列与亲代蚊体内扩增的序列比较,出现3个位点的突变,分别位于第174,215,336位点;亲代蚊体内扩增的DEN-2 E基因区部分序列与DEN-2 NGC株E基因区序列相同。结论:DEN-2在垂直传播后出现病毒E基因区的点突变。

Abstract:Objective: To study the sequence changes of envelope protein gene of dengue 2 virus after transmitted from infected parent mosquito of Aedes albopictus to its first filial generation. Methods: Geographic strains of Aedes albopictus in Xingyi city of Guizhou province were orally infected with virus DEN 2, and reared in the insectarium. The total RNA of the parents and the first filial generation was obtained and the envelope gene of DEN 2 was detected with RT PCR. Positive products were sequenced and a comparison was made between the sequences of DEN 2 isolated from infected parents and the filial generation. Results: Three point mutations of E gene were found locating in sites of 174,215,336,respectively. Sequence of DEN 2 isolated from infected parent mosquito was the same as that of DEN 2 NGC strain. Conclusion: E gene mutations might occur after the vertical transmission.
