口腔鳞状细胞癌中人乳头瘤病毒16型转化基因E_7的检测

【目的】准确检测口腔鳞状细胞癌及正常口腔粘膜中人乳头瘤病毒 16型 (HPV16)的感染状况 ,探讨人乳头瘤病毒 16型与人口腔鳞状细胞癌之间的关系。【方法】从 30例未经治疗的口腔鳞状细胞癌患者肿瘤组织及 30例健康志愿者正常口腔粘膜切取标本 ,液氮冷冻保存 ;常规提取组织DNA ,设计HPV16E7转化基因特异性引物 ,进行聚合酶链反应 ,对PCR结果进行检测 ,记录结果。【结果】 30例口腔鳞状细胞癌患者中检测到 11例HPV16阳性 ,感染率 36 7% ;30例正常对照组中检测到 4例HPV16阳性 ,感染率 11 1%。经 χ2 检验 ,两者差别有显著性。【结论】人乳头瘤病毒 16型与人口腔鳞状细胞癌有一定关系 ,但HPV16在人口腔鳞状细胞癌发生发展过程中所起的作用还有待进一步研究。

Detection of the E_7 Transform Gene of Human Papillomavirus Type 16 in Human Oral Squamous Cell Carcinoma

Objective To determine the prevalence of human papillomavirus(HPV) 16 in patients with primary oral squamous cell carcinoma(OSCC). Methods DNA was extracted from freshly frozen tumor tissues of 30 patients with primary oral squamous cell carcinoma and from the oral mucosa of 30 healthy controls. A pair of specific primers of the E 7 early gene of HPV 16 were designed. PCR products were analyzed by agarose gel electrophoresis and the results were photographed. Results HPV 16 was detected in 36 7%(11/30) of oral squamous cell carcinoma patients and 11 1%(4/30) of controls. Conclusions HPV 16 has significant association with oral squamous cell carcinoma. However, the role HPV 16 played in the tumorigenesis of oral cancer and its clinical significance remain to be investigated.
