吲哚-3-甲醇对甲醛合次硫酸氢钠致小鼠急性毒性及精子畸形率的影响

摘    要:目的确定甲醛合次硫酸氢钠(又名吊白块)半数致死剂量LD50,并了解吲哚-3-甲醇对吊白块LD50及对吊白块所致小鼠精子畸形的影响。方法采用改进寇氏法测定吊白块LD50及联合应用吲哚-3-甲醇和吊白块后并持续给予吲哚-3-甲醇,观察吊白块LD50及精子畸形的变化。结果吊白块NIH小鼠经口灌胃LD。为8711mg/kg·bw,联合应用吲哚-3-甲醇和吊白块可降低吊白块急性毒性(LD。为10000mg/kg·bw)(P〈0.05),并可显著降低吊白块致精子畸形的发生(P〈0.05)。结论根据国家标准评价,吊白块毒性分级属实际无毒级;持续摄入一定浓度水平的吲哚-3-甲醇可有效降低吊白块的急性毒性,且对低剂量吊白块所致精子畸形具有一定的预防作用。

Effects of Indole-3-Carbinol on Sodium Formaldehyde Sulfoxylate Induced Acute Toxicity and Abnormal Sperm Formation

Abstract:Objective To determine the 50% lethal dose (LD50) of sodium formaldehyde sulfoxylate (SFS) ,and the effects of indole-3-carbinol on SFS-induced abnormal sperm cell formation. Methods 50 NIH mice, fed with normal diet and water, were treated with various doses of SFS by gavage and then randomly divided into five groups. Another 50 NIH mice were fed with normal diet and 0.5% indole-3-carbinol solution for 2 days. The mice were then randomly divided into five groups and treated with 3% indole-3-carbinol solution at 1.5 hour before the administration with various doses of SFS by gavage. The signs of acute toxic symptom and death rate were recorded. The rate of abnormality of sperm cells was determined according to the method recommended by State Bureau of Standard. Results The LD50 of SFS was 8 711 mg/kg of the body weight. Results from Logistic Regression Analysis showed that pretreatment of the mice with indole-3-carbinol remarkably decreased the acute toxicity of SFS (P〈0.05). Indole-3-carbinol also reduced the degree of sperm abnormality. Conclusion According to the National standard, SFS is classified as a non-toxic substance. Long-term administration of indole-3-carbinol, a bioactive food component found in cruciferous vegetables, may protect SFS-induced acute toxicity and also reduce the rate of sperm abnormality induced by SFS.
