感染大肠杆菌后家蝇幼虫血淋巴细胞的观察

摘    要:目的 探讨家蝇三龄幼虫被大肠杆菌感染后的细胞免疫反应。方法 观察家蝇三龄幼虫感染大肠杆菌后4、8、16及24h血淋巴细胞总数(THC)及各类血细胞数量(DHC)的变化。观察大肠杆菌感染后家蝇幼虫血细胞形态的变化。结果 ①与对照组比较,感染4、8、16、24h组THC均有显著增加(P〈0.01);感染后各时间组的浆血胞和粒血胞数均显著升高(P〈0.01);感染后16、24h组的珠血胞数显著升高(P〈0.01);各时间组的原血胞和类绛血胞数量无明显变化(P〉0.05)。②感染后4、8、16h组的浆血胞和粒血胞的比例明显增加(P〈0.01),感染后各时间组的珠血胞的比例均明显减少(P〈0.01),各时间组原血胞和类绛血胞的比例均无明显变化(P〉0.05)。③感染大肠杆菌后,浆血胞发生空泡、细胞变形等变化。结论 家蝇幼虫感染大肠杆菌后在体内诱发的细胞免疫反应表现为血淋巴细胞数量、分类及形态出现变化,其中浆血胞和粒血胞两类细胞是免疫反应的主要参与者,珠血胞也可能参与细胞免疫反应。

Abstract:Objective To explore the cell immunity of the 3rd stage larva of Musca domestica after being infected by Escherichia coli. Methods The changes of the number of total hemocytes (THC), the number of the different hemocytes (DHC) and the hemocytes morphology of the 3rd stage larva of Musca domestica after being infected 4 h, 8 h, 16 h and 24 h by E.coli were observed. Results (1)Compared with the control groups, the number of THC after 4 h, 8 h, 16 h and 24 h of infection were significantly increased(P〈0.01), and the number of plasmatocytes and granulocytes of the all time points after infection were significantly increased(P〈 0.01 ), the number of spherulocytes of 16 h and 24 h after infection were significantly increased (P 〈 0.01), but the number of prohemocytes and oenocytoids of the all time points were not obviously different (P〈0.05). (2)The proportion of plasmatocytes and granulocytes of 4 h, 8 h and 16 h after infection were significantly increased(P〈0.0l), and the proportion of spherulocytes of all time point after infection were significantly decreased (P〈 0.01 ), but the proportion of prohemocytes and oenocytoids of the all time points were not obviously different (P〈 0.05). (3)The vacuolation and change in morphologies were found in plasmatocytes after being infected by E.coli. Conclusion E.coli can induce some defensive cell immune reactions in the Musca domestica larva, showing the changes of amount, classification and morphology. The plasmatocyte and granulocyte are primary participants; spherulocytes possibly participate in the cell immune reactions.
