多肽CSNRDARRC与膀胱癌细胞结合亲和性及其位点的研究

摘    要:目的研究肽段CSNRDARRC与T24膀胱癌细胞结合的亲和性及其结合位点。方法利用流式细胞仪和激光共聚焦显微镜观察多肽CSNRDARRC与T24膀胱癌细胞的结合能力以及结合位点的位置。结果流式细胞仪检测结果显示,多肽CSNRDARRC与T24膀胱癌细胞的结合呈浓度依赖性;激光共聚焦显微镜检测结果显示,肽段与细胞的结合位点位于细胞核内。结论肽段CSNRDARRC和膀胱癌细胞具有高度的亲和性且可能结合位点位于细胞核内,为进一步的膀胱肿瘤活体定位提供了一定的实验研究基础。

Abstract:Objective To research the affinity of peptides CSNRDARRC with human T24 bladder cancer cell.MethodsThe binding ability of peptides were determined by FACS and the binding sites were determined by confocalmicroscope.ResultsFlowcytometry showed that peptides CSNRDARRC bind to T24 in a concentration-dependent manner,the laser scanning confocal fluorescence microimaging system confirmed that peptides CSNRDARRC was located in Nucleus.ConclusionsPeptides CSNRDARRC have high affinity with human T24 bladder cancer cell and have the binding sites in Nucleus.This experiment improved some experimental basis and foundation for using the peptides in vivo characterization.
