羊膜复合组织片修复鼠裸露硬腭骨面的组织学观察

摘    要:目的:探讨裸露的硬腭用转染γ-IFN基因的羊膜复合组织片修复后的组织形态学特点及其抑制瘢痕形成的机制。方法:对4组(每组20只)3周龄雌性SD大鼠裸露的左侧硬腭骨面分别采取靠肉芽组织自行生长修复、羊膜移植、羊膜负载的羊膜复合组织片移植、转染γ-IFN基因的羊膜复合组织片等不同的方法进行修复,然后通过HE染色、苦味酸天狼猩红染色、偏振光显微镜及电镜检查的方法观察不同修复法分别在术后3、6、8周时各种纤维成分含量及瘢痕形成的情况。结果:术后3、6、8周,各相同时期相比,用γ-IFN基因转染的羊膜复合组织片和羊膜复合组织片修复组,其Ⅰ、Ⅲ型胶原纤维的含量明显少于其它组。结论:用羊膜负载的羊膜复合组织片及转染γ-IFN基因的羊膜复合组织片修复硬腭裸露的软组织缺损,瘢痕纤维化程度低,减少了瘢痕的形成。

Abstract:Objective: To observe the histological characteristics of resurface of denuded hard palate with oral mucous cell transfected with interferon-γ , and its mechanism in restraining cicatrix. Methods: 80 SD rats (3 weeks, female), of which mucoperiosteum of left palate were resected, were devided into 4 groups randomly. The denuded hard palate were covered with amnion transplantation(group Ⅲ), amnion loaded with oral mucous cell (group Ⅲ), amnion loaded with oral mucous cell transfected by interferon-γ(group Ⅳ). In control group, the hard palate was left denuded(group Ⅰ). 3,6 and 8 weeks later, the resurfaced tissue were examined by histology, pathology light microscopy and electron microscopy. Results: The amount of Ⅰ,Ⅱ type collagen fibrils in group Ⅲ and Ⅳ were significantly lower than other groups. Conclusion: Recovering the denuded hard palates with transplanting oral mucous cell or the oral mucous cell transfected with interferon-γ loaded by amnion, can degrade the degree of cicatrical fibrosis and decrease the quantities of cicatricles.
