摘    要:目的 探讨肥胖对肾小球足细胞的损害. 方法 以高脂喂养SD大鼠16周建立肥胖动物模型,检测尿肌酐及尿微量白蛋白,透射电镜观测肾小球足细胞的形态,抽提肾皮质总RNA并逆转录,采用荧光实时定量PCR法测定足细胞相关标志物mRNA表达. 结果 与对照组相比,高脂诱导肥胖组大鼠体质量、血脂、空腹胰岛素、空腹及糖耐量后血糖、尿微量白蛋白/尿肌酐比值均明显升高,透射电镜可见肥胖组出现足突融合现象,足细胞相关标志物Nephrin、Podocin、Podoplanin mRNA的表达量在肥胖组明显升高,而SPARC、Podocalyxin、α Actinin-4、P27在两组间差别无统计学意义. 结论 高脂喂养SD大鼠16周的肥胖模型中已出现微量白蛋白尿、足突融合以及足细胞裂孔隔膜蛋白、表面阴性电荷蛋白mRNA表达量改变.

Abstract:Objective To examine the obesity-related changes of podocyte-associated molecules nephrin,podocin and podoplanin in the kidney glomerulus of obese rats. Methods The urinary albumin and creatinine of 12 male obese SD rats fed with high fat food were monitored.The ultrastructural changes of podocyte of kidney glomerulus were investigated under electron microscopy,and mRNA expression profile of several podocyte-associated molecules was examined by real-time PCR.
