雷帕霉素在大鼠梗阻性黄疸模型中对肝脏保护作用的研究

摘    要:目的:探讨雷帕霉素对大鼠梗阻性黄疸肝脏保护作用的相关研究。方法:采用胆总管结扎方法建立大鼠梗阳性黄疸模型,将54只SD大鼠随机分为假手术组(A)组18只,梗阻性黄疸(B)18只,梗阳性黄疸+STAT抑制剂雷帕霉素RPM处理组(C)18只。分别于术后1、3、5d处死大鼠,显色基质法测定各组内毒素含量,取肝脏组织测定超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,RT-PCR测肿瘤坏死因子基因表达。结果:B和C组TNF-α mRNA表达水平均增加,B组较C组内毒素水平含量低,肿瘤坏死因子基因表达水平低,SOD活性相对高,MDA含量低,有显著差异。结论:梗阻性黄疸时TNF-α表达明显升高,通过雷帕霉素干预,肝组织TNF-α 的表达下降,SOD活性增加,MDA含量减少,可防止梗阳性黄疸所致失控性炎症反应性肝损伤。

Abstract:Objective: To study the role of Rapamycin in rats with obstructive jaundice and liver protective effect. Mehods: The common bile duct ligation in rats with obstructive jaundice model, 54 SD rats were randomly divided into A, B and C, each group of 18 rats. Respectively, after surgeryld, 3d, 5d rats were sacrificed, and the chromogenic substrate method for the determination of en- dotoxin content in each group, the liver tissue for the determination of superoxide dismutase(SOD) activity and malondialdehyde (MDA), RT-PCR, measured tumor necrosis factor gene expression. Results: TNF-alpha mRNA expression of group B and C were increased. But group B compared with group C, the level of endotoxin and tumor necrosis factor gene expression were low, SOD activity is relatively high, MDA content is low too, there are significant differences. Conclusion: TNF-~ expression in obstructive jaundice significantly higher, but with rapamycin can be further lowered liver tissue TNF-a expression and increased SOD activity, reduce the MDA content, which may prevent obstructive jaundice caused by uncontrolled inflammatory response induced liv- er injury.
