血管内皮生长因子防止脊髓运动神经元凋亡的研究

摘    要:目的研究成年大鼠脊髓损伤后血管内皮生长因子对运动神经元的神经保护作用以及Caspase-3 mRNA表达与运动神经元凋亡的关系。方法用72只SD大鼠制作成脊髓损伤模型,随机分为对照组、应用VEGF组和VEGF拮抗组。伤后6、12、24h和3、7、14d取材。应用原位杂交、TUNEL技术研究运动神经元的凋亡。结果Caspase-3 mRNA在VEGF组脊髓组织中的表达下调,在VEGF拮抗组中表达增高。TUNEL阳性细胞计数显示脊髓损伤后VEGF拮抗组有多量神经元丢失,而VEGF组神经元丢失数量明显减少。结论VEGF可以防止成年大鼠脊髓损伤后运动神经元的凋亡,其作用可能是由Caspase-3介导的。

Abstract:Objective To study the neuroprotection of Vascular endothelial growth factor (VEGF) on motor neurons after spinal cord injury in adult rats and the relationship between caspase-3 mRNA expression and apoptosis of motor neurons.Methods Spinal cord injury models were established in 72 SD rats.All rats were divided into control group, VEGF treated group and anti-VEGF treated group.The spinal cord of T13 was obtained at 6,12,24h and 3,7,14d after injury.In situ hybridization and terminal deoxynucleotidyl transferase mediated nick end labelling (TUNEL) staining were used to study the apoptosis of motor neurons.Results The expression of Caspase-3 mRNA in spinal cord was down-regulation in VEGF group and up-regulation in ANTI-VEGF group.TUNEL positive cells show that a lot of motor neurons were lost in anti-VEGF antibody treated animals, but only a few of the neurons were lost in animals that received VEGF.Conclusion These results suggest that VEGF prevented the apoptosis of spinal motor neurons in adult rats after spinal cord injury, and that effect is mediated by caspase-3.
