粒细胞集落刺激因子动员自体骨髓干细胞移植对心梗大鼠慢性心力衰竭的治疗作用

摘    要:目的探讨粒细胞集落刺激因子动员自体骨髓干细胞移植对心梗大鼠慢性心力衰竭的治疗作用。方法建立Wistar大鼠心肌梗死后心衰模型,随机分为移植组、动员组和对照组各10只。移植组于梗死区内注射经标记后的骨髓间充质干细胞,动员组皮下注射粒细胞集落刺激因子,对照组未行任何处理。4周后采用超声和生理记录仪描记左室收缩压(LVSP)、左室舒张压(LVEDP)、左室压力最大上升或下降速率(±LVdp/dtmax)等指标。取心脏组织做病理检查,观察移植细胞分化、促血管生成、细胞凋亡情况。结果细胞移植4周后,移植组及动员组心脏功能较对照组显著改善(P均〈0.05),梗死区血管密度明显高于对照组,心肌细胞凋亡指数低于对照组(P均〈0.05)。结论骨髓干细胞动员可能为慢性心力衰竭的治疗提供一种新的无创性手段。

Abstract:Objective To investigate the effect of bone marrow mesenchymal stem cells (BMMSCs) transplantation mobilized by granulocyte colony-stimulating factor (G-CSF) for the therapy of chronic heart failure (CHF) in rats. Methods CHF after myocardial infarction (MI) model was established, and then were randomly divided into transplanted group, mobilized group and control group. In transplanted group ( n = 10), BMMSCs labelled with DAPI transplantation was performed. In mobilization group (n = 10), G-CSF was injected subcutaneously. Control rats (n = 10) did not receive any treatment. Echocardiography were performed for the evaluation of cardiac function 4 weeks later, in the same time BMMSCs' s proliferation, differentiation and their angiogenic function were observed. Results Four weeks after cell transplantation, there were better heart function, higher vessel density of infarction zonein and lower apoptotic index in transplanted and mobilized group than those in control group ( all P 〈 0.05 ). Conclusions BMMSCs mobilization may offer a new and non-invasivc therapeutic strategy for CHF.
