海南红树林真菌WC1018的鉴定及免疫活性研究

摘    要:目的对从海南省文昌市清澜港红树林土壤样品中分离到的菌株WC1018进行菌属鉴定和免疫活性测定。方法分离、提纯菌株WC1018,分别采用固体平板培养法、插片培养法观察其菌落形态和显微形态特征,并采用5.8SrDNA-ITS序列测定及其系统发育分析法进行分子生物学鉴定;利用四甲基偶氮唑盐(MTT)法检测菌株的免疫活性。结果①WC1018的菌落形态和显微形态特征与曲霉属相似;系统发育树上与菌株WC1018同源性较高的序列均为曲霉属,其中与杂色曲霉菌株LTBF011-1的5.8SrDNA及ITS序列同源性最高,相似率及Bootstrap支持率均为100%。②WC1018发酵液处理后细胞增殖活性显著强于阴性对照。结论白海南省文昌市清澜港红树林土壤样品中分离到的菌株WC1018为具有显著免疫增强活性的杂色曲霉。

Abstract:Objective To identify a new strain WC1018 capabile from mangrove soil of Qinlan harbor in Hainan, and screen its immunity activity. Methods The strain WC1018 was isolated and purified, the colonial and microscopic morphology were observed using solid plate culture method and male tab culture method, the morphological characteristics were analyzed by 5.8SrDNA-ITS sequence and phylogenetic analysis. The immunity activity was assayed by MTT method. Results (1) The colonial and microscopic morphology of WC1018 were similar to aspergillus; the strain with high homology to WC1018 in the phylogenetic tree was aspergillus, the strain with the highest homology were LTBF 011-1 5.8SrDNA and ITS of aspergillus versicolor, the similar rate and Bootstrap favorability rate were both 100%. (2)The proliferation activity of the cell treated with WC1018 fermentation liquor was significantly higer that of the negative control. Conclusion The strain WC1018 capabile from mangrove soil of Qinlan harbor in Hainan was aspergillus versicolor possessing immunity activities.
