摘    要:目的: 优选腰痛宁贴膏中马钱子的渗漉提取工艺。 方法: 以士的宁转移率为指标,采用正交试验法考察乙醇体积分数、溶媒用量和渗漉速度对马钱子渗漉提取工艺的影响。采用HPLC测得士的宁含量,色谱条件为流动相甲醇-水-冰醋酸(20:96:7,三乙胺调节pH 3.10),流速1.0 mL·min-1,检测波长254 nm。 结果: 最佳提取工艺为加15倍量70%乙醇以2 mL·kg-1·min-1的速度渗漉提取,士的宁转移率>90%。 结论: 优选的提取工艺合理、重复性好,对腰痛宁贴膏的工业化生产具有指导意义。

Abstract:Objective: To optimize percolation process of Strychni Semen from Yaotongning plasters. Method: With transferring rate of strychnine as index,orthogonal test was adopted to optimize percolation extraction technology with the concentration and amount of ethanol,percolation rate as factors.The content of strychnine was determined by HPLC,chromatographic conditions were as follows:mobile phase of methanol-water-acetic acid(20: 96: 7,pH was adjusted to 3.10 with triethylamine),flow rate 1.0 mL·min-1,detection wavelength 254 nm. Result: Optimum percolation process was added 15 times the amount of 70% ethanol with percolation speed of 2 mL·kg-1·min-1.Under these conditions,transferring rate of strychnine was more than 90%. Conclusion: Optimized extraction technology was reasonable,convenient and feasible,this study had guiding significance for industrial production of Yaotongning plasters.
