川芎嗪对小鼠Lewis细胞增殖与凋亡的影响

摘    要:目的探讨中药川芎嗪对小鼠Lewis细胞的增殖和凋亡作用的影响。方法采用噻唑蓝法检测川芎嗪对Lewis细胞增殖的抑制作用,倒置显微镜观察药物作用前后细胞形态学的变化情况,流式细胞术(FCM)观察Lewis细胞凋亡指标的变化。结果川芎嗪能够抑制Lewis细胞的体外增殖,12.5、25.0、50.0 mg·L-1组的抑制率分别为29.8%、50.5%、61.3%,其48 h半数抑制浓度(IC50)为33.504 mg·L-1。实验组Lewis细胞经12.5、25.0和50.0 mg·L-1的川芎嗪处理48 h后,其凋亡率分别为(19.23±0.17)%、(30.81±0.46)%、(55.26±0.19)%;对照组为(0.64±0.37)%,实验组与对照组比较差异均有统计学意义(P<0.05)。结论川芎嗪能够抑制Lewis细胞增殖,其作用可能是通过诱导Lewis细胞凋亡和分化实现的。

Abstract:Objective To investigate the effect of ligustrazine on proliferation and apoptosis of Lewis cells of rats. Methods The inhibition of ligustrazine on the proliferation of Lewis cells was assayed by thiazolyl blue. Morphological changes were observed by inverted microscope, and cell apoptosis was detected by flow cytometry ( FCM ). Results Ligustrazine could inhibit the proliferation of Lewis cells. Inhibition rates of 12.5,25.0,50.0 mg· L-I group were 29.8% ,50.5% and 61.3% respectively. The inhibitory concentration 50( IC50 ) of ligustrazine in 48 hours was 33. 504 mg · L-1. Forty-eight hours after ligustrazine treatment, the apoptosis rates of 12.5,25.0,50.0 mg· L-1 group were ( 19.23 ± 0. 17)% , (30.81 ± 0. 46) % and (55.26 ± 0. 19 ) % respectively ; the apoptosis rates of control group was ( 0. 64 ± 0. 37 ) % , there were statistic differences of the apoptosis rates between different concentration ligustrazine group and control group( P 〈 0. 05 ). Conclusion Ligustrazine can inhibit the proliferation of Lewis cells by inducing the apoptosis and cells differentiation.
