摘    要:目的观察急性一氧化碳中毒(ACMP)后小鼠心肌酶的变化,探讨一氧化碳(CO)的心肌毒性。方法健康C57BL/6J雄性小鼠40只,随机分为对照组(腹腔注射空气80 mL·kg-1)、低剂量组(腹腔注射CO40 mL·kg-1)、中剂量组(腹腔注射CO 60 mL·kg-1)和高剂量组(腹腔注射CO 80 mL·kg-1),每组10只。腹腔注射24 h后处死小鼠,比色法检测小鼠心肌中Na+-K+-三磷腺苷(ATP)酶、乳酸脱氢酶(SDH)活性的变化;全自动生化仪检测血清心肌酶变化;心电图机检测小鼠心肌电位变化。结果与对照组比较,低剂量组小鼠心肌中Na+-K+-ATP酶、SDH活性升高,而中、高剂量组小鼠心肌中Na+-K+-ATP酶、SDH活性显著下降;与对照组比较,血清心肌酶水平随染毒剂量增加而升高,其中高剂量组心肌酶水平较对照组显著升高,差异有统计学意义(P<0.05)。心电图结果显示,与对照组比较,低、中、高剂量组小鼠心率加快,其中高剂量组部分小鼠出现心律失常。结论 ACMP会导致小鼠心肌酶水平的改变,提示临床上应重视CO中毒后的心肌损伤。

Abstract:Objective To observe the changes of myocardial enzyme after acute carbon monoxide pmsonmg( ACMP) in mice and explore the cardiac toxicity of carbon monoxide(CO). Methods Forty healthy C57BL/6J male mice were randomly divided into control group ( 80 mL·kg-1 air, intraperitoneal injection), low dose group (40 mL ·kg-1 CO, intraperitoneal injection), medium dose group ( 60 mL ·kg-1 CO, intraperitoneal injection) and high dose group ( 80 mL ·kg-1 CO, intraperitoneal injection),with 10 mice in each group. The mice were killed 24 h after intraperitoneal injection, myocardial Na + -K + -a- denosine triphosphatase(ATPase) and succinate dehydrogenase(SDH) were detected by colorimetry. Myocardial enzymes in serum were detected by automatic biochemical analyzer. The potential of mouse myocardium was checked by electrocardiogram. Results Myocardial Na + -K + -ATPase and SDH activity were increased in low-dose group while decreased in medium dose group and high dose group compared with those in the control group. Compared with the control group, myocardial enzymes were increasing as the dose of CO increasing in low dose group, medium dose group and high dose group, and there was significant difference of myocardial enzymes between high-dose group and control group ( P 〈 0.05 ). Heart rates of mice in low dose group, medium dose group and high dose group increased compared with the control group based on electrocardiogram, in which there were arrhythmia in high-dose group. Conclusion The myocardial enzymes were changing after ACMP in mice, which suggesting that cardiac injury should be considered after CO poisoning in clinic.
