一株秋茄内生真菌Fusariumsolani sp. 次级代谢产物及其抗虫活性研究

摘    要:目的 研究一株药用红树秋茄(Kandelia candela)来源镰刀菌属真菌Fusariumsolani sp. QJS4-1-2的次级代谢产物及其抗虫活性。方法 综合利用正反相硅胶柱层析、SephadexLH-20凝胶柱层析、半制备高效液相色谱(HPLC)等色谱分离方法,对真菌QJS4-1-2固体发酵产物的乙酸乙酯部位(粗浸膏)进行系统分离纯化;运用核磁共振波谱(NMR)、质谱(MS)等方法并结合文献报道数据确定化合物结构,并对其抗虫活性进行测试。结果 从秋茄内生真菌Fusariumsolani sp. QJS4-1-2发酵产物中分离得到10个化合物,包括9个香豆素类化合物orlandin (1)、demethylkotanin(2)、rubrofusarin B(3)、flavasperone(4)、6-hydroxy-8-methoxy-3-methyl-1H-2-benzopyran-1-one(5)、7-hydroxy-4-methoxy-5-methylcoumarin(6)、5-hydroxy-2-hydroxymethyl-4H-chromen-4-one(7)、3S,4R-(+)-4- hydroxymellein(8)和2-[(R)-hydroxyphenyl- methyl]-4H-pyran-4-one(9);此外还有1个生物碱类化合物aspernigrin A(10)。结论 化合物1~10均是首次从秋茄内生真菌中分得,并具有一定的抗虫活性,该研究为秋茄及其内生真菌的开发利用提供了一定的基础。

Abstract:Objective To study the secondary metabolites and insecticidal activities of the fungus Fusariumsolani sp. QJS4-1-2 derived from a medicinal mangrove Kandelia candela. Methods The ethyl acetate crude extract from the solid fermentation product of fungus QJS4-1-2 was systematically separated; and the isolated compounds were separated bynormal and reverse phase silica gel, Sephadex LH-20, semi-preparative HPLC column chromatography, thin layer chromatography and so on. And then their structures were identified by 1D NMR, MS, other spectral analysis methods and comparison with literature data, they were also tested for their insecticidal activity. Results Ten known compounds were isolated from the fungus Fusariumsolani sp. QJS4-1-2, including nine coumarin orlandin (1)、demethylkotanin (2)、rubrofusarin B (3)、flavasperone (4)、6-hydroxy-8-methoxy-3-methyl-1H- 2-benzopyran-1-one (5)、7-hydroxy-4-methoxy-5-methylcoumarin (6)、5-hydroxy-2-hydroxymethyl- 4H-chromen-4-one (7)、3S,4R-(+)-4-hydroxymellein (8) and 2-[(R)-hydroxyphenyl- methyl]-4H-pyran-4 one (9), and one alkaloids aspernigrin A (10). Conclusion Compounds 1~10 were isolated from the endophytic fungi of Kandelia candela for the first time and had certain insecticidal activity, these results provides a basis for the development and utilization of Kandelia candela and its endophytic fungi.
