MCP对结肠癌肝转移的galectin-3表达的影响

目的:探讨galectin-3在小鼠结肠癌肝转移中的表达及MCP对其抑制的作用。方法:75只Balb/c小鼠随机分为阴性对照组,阳性对照组,低浓度MCP组,中浓度MCP组,高浓度MCP组。阳性对照组和各MCP治疗组小鼠经脾脏下极包膜注入CT-26结肠癌细胞建立结肠癌肝转移模型。MCP加入饮用水中，各处理组的浓度分别为0 mg/mL， 0.01 mg/mL，0.025 mg/mL和0.05 mg/mL。3周后观察各组小鼠肝转移情况。采用酶联免疫吸附实验（ELISA）方法检测小鼠血清galectin-3浓度; 制作肝转移瘤组织芯片、用免疫组化方法检测肝转移瘤组织中galectin-3的表达。结果:（1）除阴性对照组外各组脾脏原发瘤体积中位数分别为1.51cm3，0.93 cm3，0.77 cm3和0.70 cm3。高浓度MCP组肿瘤体积较对照组明显降低（P＜0.05）。（2）除阴性对照组外，各组肝转移率依次为100%，80%，73.3%和60%。高浓度MCP组肝转移灶数目明显低于阳性对照组（P＜0.05）。（3）阳性对照组和各治疗组血清galectin-3浓度均明显高于阴性对照组（均为P＜0.01）; 阳性对照组与各治疗组间差异无显著性（P＞0.05）。（4）除阴性对照组外，各组肝转移瘤组织中galectin-3表达相互间差异无显著性（P＞0.05）。结论:galectin-3在结肠癌肝转移中呈高水平表达; MCP能明显抑制结肠癌的肝转移。

The effect of modified citrus pectin on the expression of galectin-3 in liver metastasis from colon cancer
