摘    要:目的 建立苯酚-硫酸法测定石莼多糖含量的方法.方法 用超声法提取石莼多糖,用苯酚-硫酸法进行含量测定,以葡萄糖为对照品,检测波长为480nm,用L9(34)正交试验设计优化测定条件.结果 1.0mL样液,加5%苯酚液0.8mL、硫酸5.0mL,100℃水浴保温15min结果最佳,测得石莼超声提取物中粗多糖含量为15.62%.结论 优化后的苯酚-硫酸法测定石莼多糖方法简单,重复性好,结果准确.

Abstract:OBJECTIVE To establish a phenol-sulfuric acid method to determine the polysaccharide in ulva lactuca L. METHODS Polysaccharide was obtained with the ultrasound extraction and alcohol precipitation method. Phenol-sulfuric acid method was used with glucose as parameters. The absorbance was measured at 480nm. L9 (34) orthogonal design was chosen for optimum measuing conditions. RESULTS The optimization condition was as follows:0. 8mL of 5% solution of phenol and 50mL sulfuric acid were added to 1.0mL sample solution, 100℃ water bath for 15min. The content of crude polysaccharide was 15.62% in ulva lactuca L. CONCLUSION The method after optimization is rapid with good reproducibility, simple and accurate.
