电针对局灶性脑缺血组织谷氨酸乳酸丙酮酸含量的影响

摘    要:目的:观察连续电针内关穴、曲池穴治疗7天,对局灶性脑缺血大鼠脑组织中谷氨酸、乳酸、丙酮酸含量的影响,探讨针刺治疗脑缺血的作用机制。方法:采用线栓法建立中动脉栓塞(MCAO)大鼠模型,按照神经功能评分随机配伍分为模型组、内关组、曲池组、地机组,另设立假手术组及正常组,利用韩式疼痛治疗仪电针相应穴位,7天后处死,取缺血侧脑组织行石蜡切片,HE染色观察其海马及皮质的形态学变化;酶化学法检测其谷氨酸、乳酸、丙酮酸含量。结果:电针内关穴能够显著降低MCAO大鼠缺血侧脑组织中堆积的谷氨酸(P<0.05)。电针内关穴、曲池穴能够显著降低MCAO大鼠缺血脑组织中的乳酸含量(P<0.05),同时提高其丙酮酸含量(P<0.05)。结论:电针内关穴、曲池穴能够降低脑缺血兴奋性氨基酸毒性作用,同时提高无氧酵解产物乳酸的利用率,为神经元提供能量支持。

Abstract:Objective: To investigate the effect of electroacupuncture at Neiguan and Quchi for 7 days on the content of glutamate,lactate,pyruvic acid in the ischemia brain tissue in order to provide theoretical basis for the research of the mechanism of acupuncture upon ischemic stroke. Methods: MCAO in rats were induced using suture-occluded method and according their neural function evaluation randomly divided into the model group,Neiguan group,Quchi group and Diji group,the sham group and normal group were designed as controls. All the rats were sacrificed after acupuncturing corresponding acupuncture points by electroacupuncture instrument for 7 days,and then stripping ischemic brain tissue on ice cubes. Paraffin sections and HE coloration were used to observe the histological changes in cortex and hippocampus. The content of glutamate,lactate and pyruvic acid was observed to study the effect of electroacupuncture on the ischemia brain tissue. Results: The electroacupuncture at Neiguan drastically reduced the excessive brain glutamate content in MCAO rats( P < 0. 05). Electroacupuncture at Neiguan and Quchi significantly raised brain pyruvic acid content( P < 0. 05) while drastically reduced brain lactate content in MCAO rats( P < 0. 05).Conclusion: The electroacupuncture at Neiguan and Quchi can not only reduce the excessive brain glutamate,which may mediate its role against excitotoxicity caused by glutamate,but also promote the use of the lactate to meet the energy requirement for nerve cells when the brain is short of blood. This may be one of the important mechanisms that the acupuncture can improve ischemic stroke.
