尼莫地平对脑缺血再灌注小鼠保护机制的研究

摘    要:目的 :观察尼莫地平对小鼠脑缺血再灌注损伤的保护作用。方法 :将 6 3只昆明种小鼠分为假手术组、手术组和尼莫地平组 ,后两组结扎双侧颈总动脉 ,阻断血流 30min后恢复血流灌注 ,分别给予尼莫地平治疗和生理盐水对照。检测各组手术前及手术后各时间段血小板选择素CD6 2 p、红细胞内钙(IECa2 + )、红细胞变形指数 (DI)和全血粘度 (WBV) ,并观察脑组织形态学改变。结果 :尼莫地平组术后第 1天和第 2天CD6 2 p、IECa2 + 明显高于假手术组(P <0 .0 5) ;但随着治疗天数的增加 ,CD6 2 p、IECa2 + 下降至假手术组水平。脑组织形态学病理改变以手术组较重 ,其神经细胞数量明显减少 ,残存的细胞大部分肿胀、固缩或者变性、崩解。尼莫地平组仅见少数散在神经细胞肿胀、固缩、变性。结论 :尼莫地平治疗对缺血再灌注小鼠脑有保护作用 ,有利于脑血循环、脑功能的恢复。

Abstract:Objective:To observe the protective effects of Nimodipine on mi ce with cerebral ischemia-reperfusion.Method:They selected sixty-three kunming-species mice for the trial and divided into the sham operation group,operation group and Nimodipine group.The bilateral common carotid arteries of the mice were ligted and the bloo d flow were occluded.After 30 minutes,the ligated arteries were losen and the bl ood flow reperpused.Nimoidpine was respectively given and contrasted.Then platel et-selection-CD 62p(CD 62p) intraerythrocytic-calcium(IECa 2 ) ,dyformity-index(DI) and whole blood viscosity(WBV) were examined at the time a fter operation and behind it.At last,to observe changes of the brain tissue morp hology.Results:CD 62p,IECa 2 are markedly higher in Nimodi pine group at the 1st,2nd day after operation than that in sham operation group( P<0.05).But with the days of treatment CD 62p,IECa 2 are decreas ed,It is fallen to the level of the sham operation.The morphological pathology change in brain tissues,the number of neuron cells decreased obviously in opera tion group morever,most of these residual cells became swelling,Pyknotic or dege nerative decompositive.These morphological changes are less obviously in Nimodi pine group.Conclusion:The treatment of Nimodipine have greatly protective e ffect to the brain of ischemic-reperfused mice and is beneficial to the recove ry of brain damage.
