| 摘 要： | 目的 探讨肿瘤坏死因子受体 (TNFR)基因转移对TNF杀伤膀胱肿瘤细胞作用的影响。 方法 建立TNFR逆转录病毒表达载体 ,通过转染包装细胞PA317产生完整病毒颗粒 ;膀胱癌BIU 87细胞经病毒感染后 ,检测细胞表面TNFR数量及TNF杀伤作用变化。 结果 TNFR基因转移前膀胱癌BIU 87细胞表面与TNF结合的位点数量平均为 912个 /细胞 ,经病毒感染后 ,细胞表面与TNF结合的位点数量平均为 2 872个 /细胞 (P <0 .0 5 ) ;TNF对经病毒感染的BIU 87细胞的杀伤作用较未经病毒感染者明显增强 (P <0 .0 5 )。 结论 以逆转录病毒为载体的基因转移方法可以提高肿瘤细胞表面TNFR的表达量 ,从而增强TNF对肿瘤细胞的杀伤作用。

| Abstract: | Objective To study the influence of gene transfer of TNFR on the tumorcidal activity of TNF. Methods By constructing retroviral expressin vector of TNFR and transfecting package cell PA317,the virus that can transfer TNFR was established.Then the virus was used to infect BIU 87 cell line.The TNFR number on tumor cell membrane and the tumorcidal activity of TNF in vitro were detected. Results The number of TNFR on cell membrane of BIU 87 cell was 912/cell and increased to 2 872/cell( P <0.05) after the infection. The tumorcidal activity of TNF in vitro was also enhanced ( P <0.05). Conclusions Gene transfer would enhance the TNFR expression on tumor cell membrane, the tumorcidal activity of TNF being enhanced by the gene transfer. |
