| 摘 要： | 目的 建立围绝经期性激素水平下降导致的干眼症兔模型,为进一步探讨围绝经期性激素水平下降导致干眼症的病理机制和实验研究创造条件.方法 取12只成年雌兔,分为空白组(A)和模型组(B),模型组采用双侧卵巢切除术,两组均饲养45 d,采用Schirmer Ⅰ试验观察其基础泪液分泌量、测定泪膜破裂时间现察其泪膜稳定性的改变,处死进行形态学(光镜、电镜、免疫组织化学)检测.结果 干眼症免模型建立后,模型组(B)Schirmer Ⅰ试验测量值明显低于空白组(A),泪膜破裂时间明显短于空白组(A),差异均有显著统计学意义(P<0.01);模型组(B)泪腺导管及腺泡上皮细胞中,IL-1β、TNF-α、Fas、FasL、Bax阳性表达的细胞数均明显高于空白组(A),差异均有显著统计学意义(P<0.01);模型组(B)未见TGF-β1、bcl-2明显表达,其阳性表达细胞数与空白组(A)相比较,差异无统计学意义(P>0.05);电镜示模型组(B)兔泪腺显微结构有明显损害.结论 兔双侧卵巢切除术成功首创了围绝经期性激素水平下降导致干眼症动物模型,能完整地模拟固绝经期干眼症自然病理过程,为实验研究围绝经期干眼症提供了理想的动物模型.
