缬沙坦对野百合碱诱导肺动脉高压模型大鼠血浆CRP、ET-1和NO水平的影响

摘    要:目的观察缬沙坦对野百合碱(MCT)所致肺动脉高压大鼠血浆CRP、ET-1和NO水平的影响。方法将健康雄性Wistar大鼠一次性项背部注射MCT(60 mg/kg),4周后成模,治疗组用缬沙坦20 mg/(kg·d)灌胃,持续4周达实验终点;然后经微导管介入测定大鼠肺动脉平均压(mPAP);计算右室肥大指数;分别测定血浆CRP、ET-1和NO水平。结果缬沙坦可有效降低MCT所致肺动脉高压大鼠mPAP、血浆CRP、ET-1水平和升高NO水平(P〈0.05)。结论缬沙坦可有效降低肺动脉高压大鼠肺动脉压,改善血浆CRP、ET-1和NO水平,作用机制可能与其对肺血管内皮细胞的保护作用有关。

Abstract:Objective To observe the changes of c-reactive protein(CRP),endothlin(ET) and nitric oxide(NO) in pulmonary hypertensive rats induced by monocrotaline after valsartan interventions.Methods The healthy Wistar rats were injected with 60 mg/kg of monocrotaline,after 4 weeks,the models were reproduced.The intervention group was treated with 20 mg/(kg·d) of valsartan intragastrically for 4 weeks.The parameters of pulmonary hemodynamics were monitored,the levels of CRP,ET-1 and NO were detected.Result The levels of the mean pulmonary arterial pressure,CRP,ET-1 were declined and the level of NO was increased after valsartan interventions(P〈0.05).Conclusion Valsartan can decrease the mPAP and ameliorate the levels of CRP,ET-1/NO in pulmonary hypertensive rats,the mechanism maybe related with its effects of protecting endothelial cells.
